How the intestinal barrier regulates what crosses
The gut barrier is layered: a mucus blanket, a single sheet of epithelial cells, the tight junctions sealing the gaps between them, and an immune surveillance network just beneath. Tight junction proteins including claudin, occludin, and ZO-1 govern the paracellular route, while nutrients are largely taken up through the cells themselves.
Permeability is a normal, adjustable property rather than an on-off state. The barrier samples its environment continuously while keeping most microbes and large antigens out of circulation.
What is and is not established about leaky gut
Inflammation, dysbiosis, certain medications, and physiological stress can increase permeability in experimental settings, and elevated permeability is observed alongside diseases such as celiac disease, IBD, and some infections. Whether increased permeability is a cause or a consequence in many popular wellness claims remains unresolved.
