What the C677T variant changes
MTHFR (methylenetetrahydrofolate reductase) is the enzyme that produces 5-methyltetrahydrofolate, the active folate form that feeds the methylation cycle. The common C677T variant produces an enzyme with reduced activity, especially in people carrying two copies, which can slow conversion of folate into its active form and is associated with modestly higher homocysteine, particularly when folate intake is low.
The diagram shows methylfolate entering downstream of this step, which is the basis for the claim that supplementing 5-MTHF directly bypasses a sluggish MTHFR enzyme.
Keeping the variant in perspective
MTHFR variants are very common in the general population, and for most carriers the practical effect is small, especially with adequate dietary folate. The condition has been heavily over-interpreted in wellness marketing, where it is sometimes blamed for a wide range of unrelated symptoms that the evidence does not support. Routine population testing is generally not recommended.
Methylfolate is a reasonable folate source but is not a proven treatment for conditions attributed to MTHFR. This is general educational information, not medical advice; interpret any MTHFR result with a qualified professional rather than self-prescribing.
