Voltage-gated calcium channels (VGCCs) are transmembrane proteins that open in response to membrane depolarization, allowing Ca2+ to flow down its electrochemical gradient (extracellular ~2 mM, intracellular ~100 nM at rest). This 20,000-fold concentration difference makes Ca2+ entry a powerful signaling trigger.

Once inside the cell, Ca2+ binds calmodulin, a small ubiquitous sensor protein. Calcium-loaded calmodulin activates calcineurin (a phosphatase), which dephosphorylates NFAT transcription factors, allowing them to translocate to the nucleus and drive expression of cytokine genes (IL-2, RANTES, others depending on cell type).

How voltage opens the calcium gate

Voltage-gated calcium channels (often abbreviated Cav) sit in the cell membrane and stay shut at the resting voltage. When the membrane depolarises, a change in its electrical charge, the channels open and let calcium ions flow in. Because the calcium concentration inside the cell is normally kept very low, even a brief opening produces a sharp local rise that acts as a signal.

In neurons this calcium entry triggers neurotransmitter release; in muscle it helps drive contraction; in many cells it sets off slower changes in gene activity.

From calcium spike to changed gene expression

Incoming calcium is read by sensor proteins such as calmodulin, which can activate the enzyme calcineurin. Calcineurin in turn acts on the transcription factor NFAT, allowing it to enter the nucleus and switch on genes. This chain links a fast electrical event at the membrane to longer-lasting changes in what the cell makes, a route important in learning, immune cells, and tissue remodelling.
